We are interested in the intracellular traffic and function of certain members of the low-density lipoprotein receptor (LDL-R) family. These receptors include the low-density lipoprotein receptor-related protein (LRP), megalin and ApoER2, multifunctional regarding the variety of their ligands. They can bind and mediate endocytosis of apoE and apoB bearing lipoproteins, complexes of proteases and proteinase inhibitors, complexes of vitamins A, B12 and D with their carrier proteins, extracellular matrix proteins and other proteins such as beta-amyloid precursor protein. We are currently studying their sorting signals, exocytic and endocytic pathways and the cellular machinery involved in their sorting behavior in polarized cells, using epithelial MDCK cells and neurons in primary culture as model systems. We are also studying facets of their functions in particular pathophysiological processes.
